Abstract

Hambatan degranulasi sel mast diharapkan mempercepat penyembuhan luka yang ditandai dengan meningkatnya jumlah sel fibroblas dan kepadatan sel kolagen. Ketotifen mampu mengurangi degranulasi sel mast dan mengurangi pelepasan histamin, protease sel mast, myeloperoxidase, leukotriens, PAF dan macam-macam prostaglandin, juga menghambat agregasi polimorfonuklear serta mengurangi respons inflamasi dan mempercepat migrasi fibroblas di fase proliferasi. Penelitian ini true eksperimental laboratorik dengan desain randomized controlled trial, bertujuan untuk mengetahui perbedaan jumlah sel fibroblas dan kepadatan sel kolagen pada tikus Wistar yang diberi ketotifen oral dosis 0,3 mg/kg dibandingkan dengan plasebo pada penyembuhan luka insisi. Disimpulkan bahwa ketotifen meningkatkan jumlah sel fibroblas dan kepadatan sel kolagen pada penyembuhan luka insisi tikus Wistar.

 

Inhibition of mast cells degranulation will accelerate wound healing process, indicated by increased density of fibroblast cells and collagen. Ketotifen inhibit the degranulation process and decrease releases of histamin, mast cells proteases, myeloperoxidases, leukotriens, PAF, and various prostaglandins. Ketotifen can also inhibit polymorphonuclear cells aggregation, increasing the rate of fibroblast migration in the proliferation phase. This experimental laboratory study was to identify the effects of ketotifen on fibroblast cell count and collagen density in Wistar rat’s model compared to placebo. Ketotifen has been shown to increase fibroblast cells count and collagen cells’ density in wound incision healing on Wistar rats.
